The synthesis of complex glycans by most glycosyltransferases (GTs) is a multistep process requiring a combination of both donor and acceptor substrates. The flexibility of the GTs to use a variety of glycan substrates, each with distinct structures and stereochemistry, makes the assay of GT function challenging, and yet, essential. Here we describe the development of a cell-free protein synthesis-based assay for the detection of GTs, taking advantage of the modularity of protein synthesis to assemble a complex glycan structure in a single reaction tube. The resulting glycoconjugate is then purified and quantified using a facile assay employing a commercially available HRP conjugate. A GT identified as having a glycosylation defect was tested and shown to correctly assemble the glycan structure and display the correct glycosylation phenotype in vitro, whereas a control GT displaying the same domain was found to be correctly glycosylated. This novel approach provides a rapid, scalable, and sensitive method of investigating GT defects.It seems that Colorado Sen.
